Myopathology has dysmetabolic and inflammatory disorders in pathogeneses incl. toxic cardiomyophathy and cardiomyophathy in the case of patients obesity. Plant originated products remain today an undiminished source of new pharmaceuticals. For the present study the polyphenolic fraction was isolated from bark of grey alder grown in Baltic region. The major component (>80%) of this fraction was identified as oregonin, 1,7-bis-(3,4-dihydroxyphenyl)-heptan-3-one-5-O-_-D-xylopyronoside for which. Very high antioxidant and radical scavenging activity was found. In 2 series of in vivo experiments with Vistar line rats it was shown that under conditions of induced hypercholesterolemia, application of oregonin provided high level of myocardial muscle renovation. The morphological studies revealed quickly-regenerated cells, restoring of proteins, lipids and carbohydrates metabolism. Oregonin application extensively decreased degradation of animals cardiomyocytes resulting from chronic alcohol intoxication: cells with damaged myofibrillar apparatus practically were not observed. Statistically significant elevation of tyrosine, methionine, alanine, _-alanine, asparagines and proline pool was detected in cells of myocardium. Rising of pool of amino acids, including proteinogenic ones, and non-essential/essential amino acids ratio indicated activation of protein synthesis. In an in vitro experiments with human blood, the activity of phagocytes was researched by monitoring of concentration changes of cytokines: IL-1_, IL-10, IFN-y, IL-6, TNF-_ in cells culture (GE Health Care Bio-sciences AB, Sweden) from healthy volunteers - blood donor bank (Wallenberg Lab), using multiplex "Elisa" kit. Inflammation was induced by standart Lipopolysacharide activator. After incubation with oregonin (0,025 mg/mL) the significant cytokines decreasing was detected in cells culture that indicated high anti-inflammatory activity of the diarylheptanoid.Our data reflects normalization of protein metabolism in myocardium in hypercholesterolemia, increasing of adaption and stimulation of metabolic response to alcohol intoxication after application of oregonin. This correlates with its anti-inflammatory properties detected. Therefore, oregonin can be considered as an efficient economically available component for development of pharmaceuticals for cardiomyophathy treatment.
