Communication n° 99. Serum response Factor (SRF) is a muscle-enriched transcription factor that plays an important role in the regulation of contractile protein gene expression in mammalian heart. Strong SRF expression was observed in the myocardium of the mouse heart. We adopted a conditional gene inactivation scheme for SRF by using the Cre/loxP system. We have also generated and further characterized one transgenic line in which Cre recombinase expression is under the control of the desmin promoter. A stable transgenic line (Des-Cre) was obtained and characterized by crossing with the CAG-CAT-Z reporter transgenic line. This line produces beta-galactosidase in the cells with Cre recombinase activity. A regionalised blue staining could be observed in a part of the left ventricle of the heart. We produced mice with impaired SRF expression at the same region of the heart. The histology of the Des-Cre Sf/Sf mutant was analysed from new born and adult mice. Des-Cre Sf/Sf mutant developed localised lesions in left ventricular wall resembling infarct myocardial lesions. Severe fibrosis and disorganisation of the left myocardial wall was observed with variable degrees of ventricular hypertrophy. The results suggest a cell autonomus role of SRF in adult cardiac function.
