Vertebrate skeletal muscles derive from transitory mesodermal structures called somites. Within the dorsal part of the somite, the dermomyotome, there are progenitors that express the transcription factor Pax3. The dermomyotome gives rise to the derm of the back, all skeletal muscles of the body and also contribute to endothelial and smooth muscle cells of blood vessels and brown fat. We had previously shown that reciprocal repression between Pax3 and Foxc2 genes affects cell fate decisions in these multipotent cells of the somite in which both genes are expressed. Pax7 and Foxc1 genes are also potentially implicated in this mutual inhibition and we are currently investigating conditional Foxc1/Foxc2 mutant mice, with particular reference to brown fat formation. Notch signalling is a candidate in this respect. It is known to promote the maintenance of the progenitor cell pool within differentiating skeletal muscle masses. We are investigating the role of this pathway in myogenesis by using a mouse model in which a sequence encoding the intracellular constitutively active form of Notch (NICD) was inserted into an allele of Pax3. Pax3NICD/+ embryos present defects in appendicular muscle development and trunk myogenesis. Ongoing work focuses on whether activation of the Notch pathway leads to changes in cell fate decisions of multipotent Pax3 positive progenitors in the context of the equilibrium between Pax3/7 and Foxc1/2.
